Today, the process of sperm cryopreservation serves as an integral part of reproductive technology and represents a valuable clinical assistance method in infertility management and sperm bank formation. Sperm cryopreservation is currently the most valuable and widely used method of protecting reproductive function in men undergoing gonadotoxic treatments such as chemotherapy or radiation therapy. In this study, 15 samples of normal human sperm collected from 15 men referred to the Infertility Research Center were divided into three groups: the first group of fresh samples, the second group of samples frozen commercially with frozen sperm alone (control), and the third group was frozen with the hand-made freezing medium. Then total and progressive sperm motility and sperm viability rate were measured according to WHO criteria in the study groups. Significantly improved sperm parameters and increased survival rates were observed in the hand-prepared sperm freezing medium. It can be concluded that the new freezing medium during the freezing-thawing process is an excellent strategy to increase the survival rate and increase the motility of human sperm.
