Liposomes act as vaccine and adjuvant delivery systems due to their ability to deliver drugs. The unique physicochemical properties and flexibility of liposomes in size, charge, bilayer stiffness, and compositions allow for targeted antigen delivery through a wide range of administration routes. In the development of liposomal adjuvants, the type of immune response promoted by their physicochemical properties is related to the size and charge of liposome particles that affect liposome biodistribution, exposure to lymph nodes, and innate immune system recruitment. Adding immunostimulating agents can strengthen their immunogenic properties. In this review, we examine the features that should be considered in the design and manufacture of liposomal adjuvants for vaccine delivery.
